Scientists have long studied the brain’s cells, genes, and proteins, but the role of lipids has remained largely obscure. These fatty molecules form the membranes around neurons and provide the myelin insulation that protects nerve fibers. Despite their importance in cell communication, mapping where specific lipids reside in the brain has proven difficult with conventional imaging tools.

A team at the Ecole Polytechnique Fédérale de Lausanne (EPFL) has now filled this gap. Led by Giovanni D’Angelo, Luca Fusar Bassini, and Gioele La Manno, the researchers produced a comprehensive map of lipid distribution in the mouse brain. Their work, described as a lipid brain atlas, reveals that these molecules are arranged in highly organized patterns that correspond to specific anatomical structures and chemical boundaries.

    539 Distinct Chemical Territories

To construct the atlas, the team used mass spectrometry imaging on thin sections of brain tissue. A laser scanned each slice to identify which lipids were present and where they were located. The researchers analyzed 172 different lipids across 109 brain slices from 11 mice. This process generated approximately 7 million measurements, which were combined into a 3D reconstruction using custom machine learning tools.

The resulting map identified 539 chemically distinct territories, which the team named “lipizones.” Each zone has a unique combination of lipids, creating a recognizable chemical signature. In many cases, these signatures were so distinct that researchers could determine the origin of a brain tissue sample simply by examining its lipid composition.

“In effect, lipids work like postal codes, giving distant but related parts of the brain the same chemical address,” D’Angelo said. This chemical organization often revealed relationships between nerve cell bodies and distant regions connected by fibers, connections that were difficult to detect using maps based solely on gene activity.

    Pregnancy and Brain Chemistry Shifts

The atlas also uncovered significant chemical variations in areas previously thought to be uniform. White matter, which contains the nerve fibers connecting different brain regions, was found to contain numerous distinct lipid zones. This suggests that the cells producing myelin are far more chemically varied than their genes alone would indicate.

Researchers also detected unexpected chemical boundaries in the choroid plexus, the structure responsible for producing cerebrospinal fluid, as well as in the tissues lining the brain’s internal cavities. These findings demonstrate that lipid mapping can reveal features of brain organization that are not apparent from anatomy or genetic information alone.

Beyond static structure, the atlas provided new insights into how brain chemistry changes during pregnancy. When the team examined pregnant mice, they found substantial shifts in lipid composition across numerous brain regions. These changes were more pronounced than the typical lipid differences observed between male and female mice.

The cortex, the brain’s outer layer, showed particularly extensive changes. Meanwhile, levels of galactosyl ceramide, a key component of myelin, increased sharply throughout much of the brain’s white matter. La Manno noted that these shifts suggest the brain retunes its own wiring to meet the demands of pregnancy, constantly adjusting its lipids to support the body’s changing needs.

    Implications for Human Disease Research

The Lipid Brain Atlas is freely available online, allowing scientists worldwide to explore this aspect of brain biology. While the specific map is based on mouse tissue, the method used to create it can be applied to human studies. Fusar Bassini emphasized that what transfers to humans is not the map itself, but the method and the confirmation that a healthy brain has a highly organized lipid structure.

This reference for healthy mouse brain tissue will help scientists investigate how disruptions in lipid distribution might contribute to brain development, aging, and disorders such as Alzheimer’s disease and depression. By combining lipid information with existing genetic and cellular maps, researchers can gain a more complete understanding of how the brain is organized and how its chemistry changes over time.
