    Scientists at the University of Utah Health have identified a specific mechanism by which bacteria detect and respond to viral attacks. The discovery reveals that bacteria use an enzyme produced by the invading virus itself as a warning signal. This finding provides a clearer understanding of bacterial immunity and could help researchers design more effective treatments for antibiotic-resistant infections.

    The research focuses on a class of viruses called bacteriophages, or phages. These viruses naturally infect and kill bacteria. Because they target specific microbes without harming human cells, phages are being studied as an alternative to traditional antibiotics. However, bacteria have evolved complex defense systems to survive these viral attacks. Understanding how these defenses work is essential for developing reliable phage-based treatments.

    Viral Enzyme Triggers Bacterial Alarm

    The study centers on a bacterial immune pathway known as CBASS. When activated, this system can trigger a “last resort” response. The infected bacterium kills itself before the virus can replicate and spread to neighboring cells. This self-destruction is a fatal strategy, so the system must distinguish between genuine threats and false alarms with high precision.

    Sam Hobbs, an assistant professor of biochemistry at the University of Utah Health and the lead author of the study, described the discovery as a significant breakthrough. The team found that the detection process relies on a viral protein called a protease. This enzyme is produced by certain phages to degrade other proteins. Instead of just harming the host, the protease cuts a crucial sensor molecule inside the bacterial cell. This damage serves as the alarm that activates the CBASS pathway.

    This mechanism differs from other known antiviral immune systems. Some pathways detect viral genetic material directly. In the CBASS system, the trigger comes from the activity of a viral protein acting on a protein belonging to the bacterial host. Hobbs noted that this is a totally new mechanism for how these host proteins are activated.

    Evolutionary Link to Human Immunity

    The findings have implications beyond bacterial infections. The CBASS pathway is related to an immune system found in humans. This suggests that the basic components of this antiviral defense have existed since the common ancestor shared by bacteria and humans. The pathway has been maintained for billions of years, indicating its importance in fighting viral infections across different forms of life.

    Bacteria offer a useful model for studying these fundamental immune processes. Their rapid life cycles allow scientists to observe immune responses quickly. Insights gained from bacterial studies can then be tested in biological models that are more closely related to humans. This evolutionary connection provides a window into the core mechanisms that maintain the ability to fight viruses.

    The research was published in the journal Science. The work was supported by several organizations, including the Pew Biomedical Scholars program, the Burroughs Wellcome Fund, and the National Institute of General Medical Sciences. The findings provide a concrete target for future research into overcoming bacterial resistance to phage therapy.
